Abstract:

In this study, formulation of sustained-releasingmatrix tablet of bupropion 150 mg, using hydroxypropylmethylcellu lose(HPMC) 4000cps was evaluatedwith the aim of reducing the frequency of daily dose. The level of HPMC4000 ,polyvinylpyrolidone(PVP) and magnesium stearate(Mg St)was varied based on a 2level 3 factor factorial experimental designusing the release rate of the drug from the matrices as the response variable. Themechanism of drug release from hydrophilic matrix tablets is complicated but it is known to be related to dissolution of drug and its diffusion through the hydrated portion of matrix and erosion of the outer hydrated polymer on the matrix surface.Granules of the optimum formulations were compressed into tablets using EK-O lorsch single punch tablet machine. Evaluation of tablets including weight variation, crushing strength and friability demonstrated acceptable results.Based on dissolution data of the eight tablet formulations resulted from the experimental design, a polynomial regression equation was generated and used for obtaining the optimum formulations.Invitro dissolution tests also revealed sustained release of drug for an 8 hours period at the end of which almost complete release was achieved.According to release studies formulation A,AB and C has been selected for long term stability studies.

Preparation and In vitro Characterization of Alprazolam Extended- Release Tablets Using HPMC 4000cps

The main aim of this study was preparation and evaluation of extended - release system ofthe anxiolytic substance. Alprazolam is a short-acting benzodiazepine with general propertiessimilar to those of diazepam. Our studies focused on development of extended drug deliverysystem based on Hydroxy Propyl Methyl Cellulose (HPMC 4000cps) as retard agent andPolyvinylpyrrolidone (PVP k30) as binder us...

Formulation and Evaluation of Sustained Release Matrix Tablets of Glipizide

The purpose of this study was to develop a new monolithic matrix tablet to completely deliver glipizide in a zero order manner over a sustained period. Two approaches were examined using drug in a formulation that contain polymer like hydroxylpropyl methyl-cellulose K 100 (HPMCK) and Eudragit L 100. The granules were prepared by wet granulation method and thereby formulated as F-1, F-2. F...

Formulation and Release Behaviour of Sustained Release Ambroxol Hydrochloride HPMC Matrix Tablet

Monolithic matrix tablets of ambroxol hydrochloride were formulated as sustained release tablets employing hydroxypropyl methylcellulose polymer, and the sustained release behaviour of the fabricated tablets was investigated. Sustained release matrix tablets containing 75 mg ambroxol hydrochloride were developed using different drug polymer ratios of hydroxypropyl methylcellulose. Tablets were ...

Effect of Formulation Variables on Phenobarbital Release from HPMC Matrices

Controlled release swellable tablets of phenobarbital were prepared by a simple direct compression process using hydroxypropyl methylcellulose (HPMC) as the matrix former. The effects of the viscosity grades of HPMC and HPMC/lactose ratio and ethylcellulose (EC)/sodium carboxymethylcellulose (NaCMC) and their concentrations on the release behavior of phenobarbital from HPMC matrices wer...

Formulation development and evaluation of Diltiazem HCl sustained release matrix tablets using HPMC K4M and K100M.

The aim of this study was to develop a sustained release hydrophilic matrix tablet of Diltiazem HCl and evaluates the effect of formulation variables (e.g. lubricant, binder, polymer content and viscosity grades of HPMC) on drug release. Twelve different formulations (F1-F12) were prepared by direct compression. The results of the physical parameters and assay were found to be within the accept...
